Towers are an excellent source for "Essence" and sometimes have additional rewards on harder tower floors. 

Access

You can access towers under the tab "Towers" when you click the adventure button. Towers unlock on a specific adventure rank. 

Climbing Towers

Each Tower consists of multiple Floors that have to be completed to climb further. You can come back at any time to attempt the next floor. Your progress will be saved.
